Sexual offense is a wide-ranging term used to categorize any unlawful behavior involving sexual acts, including rape and sexual assault. Detection of semen in clothing items received at the forensic laboratory is considered crucial as it may aid in identifying the semen donor through DNA analysis. Therefore, utilizing a reliable and robust method in detecting semen stains is a paramount in aiding the conviction of offenders and in some cases the exoneration of the innocent. Over the years, various techniques have been devised for screening and confirming evidence, however some have limitations. A recent increase in the number of Forensic examinations of rape victims has made improvements to existing forensic methods to increase the likelihood of detecting and recovering semen a welcome development. The National Institute of Scientific Police (INPS/LPS69), France collaboration with AXO Sciences, launched STKĀ® Sperm Trackerā„¢ an advanced technology developed to detect semen on sexual assault evidence items. The technique is reported to be highly specific and can use as a confirmatory test for semen. Indeed, the technique is rapid, sensitive, environmental friendly and easy to use. The research conducted focused on comparing its performance with traditional methods on semen identification, effect of STK on subsequent techniques such as DNA analysis, effect of STK on detecting semen in vasectomized person and its sensitivity in detecting semen on fabric and degraded semen stains. Research study revealed the suitability of STK over conventional Acid Phosphatase test, ability to use in downstream applications and semen from vasectomized donors. Furthermore, higher sensitivity on detecting semen on different fabric types and on degraded semen. These findings provide that the STK sperm tracker can be used in forensic laboratories with enhanced reliability, replacing classical methods on localization the presence of semen in evidence items.
